I have a 72 dart w/360 stroker punched out to a 408, ballanced rotating assembly, .050 cam, performer intake and a 750 eldebrock carb, electric fuel pump with pressure regulator.

Problem #1: hesitation at take off... i have adjusted the pump, trying all three positions w/ no change, played with the timing, it is at 10% initial.

other symptom: fuel drains down, filter is dry when not running. when running fuel "bubbles" in filter???

possible problem? gas tank is not vented (charcoal canister removed in build out)...strong suction when taking off gas cap...

try running the car with the gas cap off if you removed the charcoal canister and blocked of the line that could be your problem /you may have to drill a vent hole for the cap or get a vented cap from an auto parts store
 
Get your venting problem fixed first and install a fuel psi guage if you don't already have one.

I thought I had a venting problem on my car but it turned out to be a cracked fuel hose by the tank that was only 4-5 yrs old. It didn't leak gas but would suck air into the fuel line.

I swapped to stronger fuel injection hose back and front before the pump.
 
Don't know what you have for a cam but if it's much hotter than stock you will need more than 10 degrees initial timing, likely 15-20 degrees. Also, if you have the vacuum advance connected to manifold vacuum (should be on ported/timed) the loss of manifold vacuum and subsequent loss of timing when you first get on the gas may be your problem.
