Can this be done to a 440?

There are no coolant passages on the intake flange face of BB heads. If you're referring to the port in the center of the head, at the intake face, thats the exhaust transfer port (usually carboned off) that allowed hot exhaust to travel across the intake manifold and heat the air fuel charge (there's a physical wall seperates them) for winter operation. Blocking the passage is fine and recommended.

That manifold in the link you posted is for a Small Block.
