I have the new roof skin finally welded up on my 72 dart and getting ready to grind down some of the welds, I have already spot welded the roof to the quarter panel where I drilled out holes previously to de-skin it, my question is should I try to weld the seam solid or will it be fine the way it is, I'm just worried about flex and cracking filler or paint.

Full weld is overkill... The spot welds held for 40 years man. Build the gap with some short strand fiberglass, skim it with light weight, and call it a day. Also add some 2x3 sub-frame connectors, that body isn't going anywhere...

not so concerned with the spot welds holding or not but more about the sheet metal flexing between the spot welds and cracking paint

All-Metal or a similar filler over the seam, then body filler as a skim coat. I'd make sure the outside ends of the panel are fully welded if you don't want to do weld it solid.
