On a 65 Barracuda ( 273) the shifter cable goes into the transmission through a box bolted on the tranny ( i think 5 bolts). My question is this, is the only way to re clip the cable is through the little hole on the bottom ( of the box )or is there another easier way?

that is the park brake cable,(box at rear of tranny) the shift cable mounts in the front driver side, it has a locking bolt with a adjustment wheel.
65 has some 1 year only items-- do some searching on push button tranny/ cable shift. Lawrence

65 Barracuda has the shifter on the floor, not push button. The cable I'm talking about goes from the floor shifter to the small box that mounts to the bottom of the drivers side tranny.

Check your Factory Service Manual. My '65 Plymouth FSM has a pretty complete step-by-step section on the floor shift,
with the two cables, backup light switch, etc. The rest of the auto trans section goes on to show and explain how to install
and remove the cables. From my personal experience I can tell you that it's a LOT easier to deal with the shift cable if you remove that park cable cover that you referred to as a box with 5 bolts. (Drain the tranny first!)

If you don't have a Factory Service Manual, the '65 Plymouth manual I have is available here:
