I have a 727 that has the emergency brake on the tail shaft. Can that just be taken off and used in a car or will the tail shafts need to be changed?

It's a very different animal. Too much would have to be changed, in my opinion.
To use it a car you have to change the extention housing and the output shaft, not that big of a deal. The truck trans will probably have 4 pinion planetaries and 4 disk clutches in there too. They changed the spline angle in the seventies so make sure to get the right shaft, they wont interchange.
You CAN, but as Mr. Lusk eluded, it is more work than necessary when you can change to a car tail shaft and housing. The transmission itself is probably heavy duty.
