Tune up question

After I rebuilt my carburetor, is it possible to get it all dialed in with the power boost line plugged?

I sent the booster and master cylinder out for overhaul and while they are at the shop I went ahead and rebuilt my carb at home.

I’m ready to install the carb and finalize tuning it but not sure if just plugging PB line will give me erroneous vacuum readings while dialing in.

Yes the brake booster is just a resivour it does not have a metered leak or anything like a pcv valve does.

Just cap it off and tune away.
