Can I use regular compression fittings in Trans cooler lines our do they have to be double flared?

I don't know why not. I have one in my line and it doesn't leak.

Never use them in a brake line though.
 
My homemade trans cooler lines have compression unions at approx' mid length.
Never had a problem with them.
 
I'd be concerned about metal fatigue at the edge of the ferule if the line vibrates, but that would be a long way down the road.
 
compression unions are fine for everything but brake lines, heck I've even used them to repair the power steering pressure lines on cars, they normally have 1000 PSI or more on those
