would a 727 big block transmission form a truck fit in a 72 plymouth duster?

You need to see if it's 2dw, an dalso if the tailshaft has the retaining clip access port is on the side of it (pass side) or under the transmission mount (bottom). If it's on the side, you'll have to clearance the floor or change the tailhousing to a std pass car one.

its a 4x4 727 transmission

It would probably be worth grabbing for the internals, but get a passenger car mainshaft and tailhousing.
